The retailer has revealed it will host the event in aid of the game's unconventional Wednesday release.
It will take place at the firm's Stratford store, which will reportedly be transformed into a "scenic replica of the game's winter wilderness, complete with Redcoats who will be patrolling and entertaining the queue."
Meanwhile, additional midnight launches will take place across the UK, whilst Ubisoft's developer Tim Browne will attend the game's launch in Hull from 9:30pm in order to chat with customers.

?We are bringing a piece of the adventure to life at GAME Stratford for London fans and we are delighted that developer Tim Browne will be attending GAME Hull, giving both the north and south a great experience to look forward to.?
Midnight lanch events remain one of the more powerful tools retailers have in enticing gamers to purchase their games at retail and GAME will certainly be hoping this to be the case, offering a themed evening, along with the opportunity to meet with the personnel behind the game.
Assassin's Creed III will launch in the UK on Wednesday October 31st and will see GAME stores open their doors from 11:45pm on Tuesday October 30th.
However, PC gamers will be forced to wait until November 21st for the latest offering in the long-running franchise, as the title's PC version will suffer from the often delayed PC release, something of which PC gaming veterans have become accustomed to.
